WP1 : Ethics Requirements
Leader: IDE R&D
M1-M48
The objective of this workpackage is to ensure compliance with the “ethics requirements” set out in this workpackage.
WP2: Microbial collections screening
Leader: MATIS
M1-M30
This WP focuses on developing and applying a range of high-throughput approaches for siderophore and biosurfactant biodiscovery. The objectives of this work package are to assemble provided diverse microbial collections; to select and optimise culture-based and culture-independent methods for screening procedure; to identify and explore taxonomical groups with active production of for biosurfactants and siderophores; and to identify the genetic elements in charge of the synthesis of targeted biosurfactants and siderophores.
WP3: Databases integration and Industry-driven designs
Leader: IDE R&D
M1-M48
This work package combines activities aimed to obtain genetic combination designs to produce new tailor-made biocompounds. Specifically, WP3 aims to:
the development of machine learning algorithms to survey databases and collect pathways.
genetic information, molecular structures and properties of biosurfactants and siderophores.
the definition and construction of biosurfactant and siderophores chemical space.
the identification of major molecular structures with interesting properties.
the implementation of inverse QSAR/QSPR modelling approach.
WP4: Systems metabolic engineering for microbial platform optimisation
Leader: USE
M4-M46
The aim of this WP is to engineer bacterial strains with the capacity of overproducing new chemical designs. The specific goals include:
adaptation and optimisation of standardised vectors in extremophilic model organisms
the development of “ready to use” microbial strains and model-driven strain designs
experimental validation of metabolism and gene expression model.
WP5: Lab-scale fermentation and chemical characterisation of compounds
Leader: UoA
M6-M40
In this WP protocols for production of small-scale quantities, from strains derived in WP1 and WP4 will be accomplished together with isolation and characterisation of biosurfactants and siderophores.
Evaluation of the state of the art on extraction and purification procedures.
Standardised protocols for chemical characterisation.
Determination of novel biosurfactant and siderophore chemical.
WP6: Pilot systems for process optimisation
Leader: BBEPP
M12-M48
The high-performing strains developed in WP5 will be used during the optimisation of the operating strategy at pilot scale. The main objectives are:
- Integration of unit operations in mathematical process model.
- Standardised protocols for chemical characterisation.
- Core processes demonstration at pilot scale.
WP7: Proof of concept for end users’ applications
Leader: SE
M12-M48
The overall goal of this WP is to characterise, analyse and investigate the efficacy of the identified bio-surfactants in the different end-user fields (oncology, gene delivery, cosmetic, house-hold and personal care, food industry/nutrition, agriculture) of the present consortium. WP7 includes the following methods and tests:
- Integration of unit operations in mathematical process model.
- Bio-surfactants drug encapsulation, efficacy and toxicity profiles.
- Novel encapsulation approaches for different end-user applications.
- Immunomodulatory and anti-tumorigenic capacity of selected compounds.
- Efficacy of bio-compounds as chelating agents.
